This week’s overlooked batch is California science first: enzymes reading letters evolution never wrote, batteries that work better when you squeeze them, and a 1970s molecule that makes mice burn fat without eating less.

- One-line: UC San Diego showed that bacterial RNA polymerase can accurately transcribe a synthetic eight-letter “hachimoji” DNA alphabet.
- What happened: All known life uses A, T, C, and G. Chemists had already built four extra letters — P, Z, B, and S — that keep Watson–Crick geometry. Dong Wang’s group at UC San Diego’s Skaggs School of Pharmacy used biochemistry plus cryo-electron microscopy to watch E. coli RNA polymerase read a template containing all eight and write the matching RNA. The enzyme used many of the same structural cues it uses on natural bases; the P:Z pair ran about twice as slow as G:C. This is still a test-tube result, not a living eight-letter cell. 2. Squeeze the battery, don’t fight the dendrites
- One-line: SLAC and Stanford found that compressing a solid electrolyte makes lithium dendrites grow sideways instead of shorting the cell.
- What happened: Solid-state batteries fail when needle-like lithium dendrites punch through the ceramic electrolyte. Using SLAC X-rays, the team showed those needles start at internal pores and grain-boundary junctions, not only at the surface. They fitted a shape-memory-alloy ring, heated it to 170°C so it shrank, and applied a steady side-squeeze. Dendrites still formed, but they ran horizontally and never reached the electrodes. Compressed cells survived thousands of cycles, including extreme charge rates in the related Nature work. Compression becomes a design knob instead of another chemistry fight.

3. A 1970s molecule makes mice burn fat without eating less
- One-line: Berkeley’s Näär lab revived TOFA, an old lipid-synthesis inhibitor, and watched obese mice lose fat, keep muscle, and spend more energy.
- What happened: GLP-1 drugs work the “eat less” lever. Anders Näär’s group went after the other one. In Science Advances (online Aug. 21; campus coverage this week), TOFA blocked production of cholesterol and triglycerides and turned up fat-burning genes. Food intake, activity, and body temperature did not change; whole-body energy expenditure rose as much as 18 percent. Mice lost about 18 percent of body weight from fat, with lean mass largely spared, and markers of fatty liver and glucose control improved. Combined with semaglutide or tirzepatide, the effect stacked. The compound was first studied in the 1970s and never fully tested for obesity. The team has spun out ReRx Therapeutics; no human data yet.

- One-line: Mark Farver built a RANS S-21 in Mendocino County, wired and painted it himself, then flew 49 legs to Wittman Field.
- What happened: Farver spent two and a half years building N319PG — an all-metal taildragger on tundra tires, silver over navy with safety-orange trim — at Little River, California. He flew the first flight. With his wife Carin Berolzheimer calling traffic, he put it on the yellow dot at Oshkosh on July 18 after 48 days and 49 legs. 5. Silent key: Harold Kramer, WJ1B, who ran ARRL’s engine room
- One-line: The former ARRL chief operating officer and QST publisher died August 31; the notice makes a broadcasting-and-ham life visible.

- What happened: Christopher Voigt’s biological-engineering group engineered strains that act as transistors and demonstrated logic operations — multi-input, OR, IMPLY — depending on where a strain sits in a circuit. Five strains are enough, they say, to build the usual architecture pieces. Each calculation takes about eight hours, which is useless for phones and useful for plants: the intended next step is circuits on leaves or roots that sense and respond to stress. Second pass, same week. This batch is more hands-on: a Stanford circuit that turns vanishing proteins into light, a boat that flies ten feet over Narragansett Bay, and a World War II blimp hangar on the Oregon coast that just found a new steward.

Why this is the cover: It is the week’s cleanest piece of lab ingenuity — the signal gets brighter as the target disappears — and it is specific enough to open a digest.
1. COVER STORY — Protein loss, turned into a brighter signal
- One-line: Steven Banik’s Stanford lab built a genetic circuit that converts the disappearance of a protein into a louder transcriptional output.
- What happened: Targeted protein degraders are hard to screen because a small drop in a scarce protein is almost invisible. In a Cell paper published online Sept. 1, Banik’s group describes ratiometric transcriptional activation: fuse the protein of interest to a molecular brake that holds a transcription factor off; as the fusion is degraded, the brake goes with it and the “gas pedal” drives fluorescent protein, cell survival, or a sequencing barcode. 2. Ultrafast electrons, then an unexpected optical kick
- One-line: SLAC and Stanford used megaelectronvolt electrons plus a laser and saw a much stronger optical response to ionization than expected.
- What happened: SLAC posted the news brief on Sept. 3. The team fired ultrafast electrons and a laser into semiconductors and watched how the material answered optically after ionization. The response was stronger than the usual models predicted. The practical claim is modest and specific: better routes to imaging and sensing that sit between electron probes and light. This is adjacent to the same lab’s attosecond X-ray movie work at LCLS, but the Sept. 3 note is the electron-plus-laser result.

3. A seaglider leaves the water
- One-line: REGENT Craft’s first manned electric flying-boat prototype flew 1,956 feet over Narragansett Bay on Sept. 2.
- What happened: Two crew lifted about ten feet off the water and stayed airborne for 30 seconds. REGENT’s vessels dock like a monohull, rise onto hydrofoils, then fly 30 to 60 feet up in ground effect. The company says computers hold altitude so the craft can be driven like a boat, without a pilot’s license or full aviation rules. They had been foiling the 12-passenger prototype since last summer; this was the first time the full-size passenger hull flew. A $240 million round closed days earlier. A 255,000-square-foot factory is going up at Quonset.

Today, Sept. 5, EBPCO ran a four-and-a-half-hour black-and-white refresher at the 13th Street community darkroom: film process, print, instructor at the trays. 8. PVC in, engine oil out
- One-line: Virginia Tech, with Texas A&M and Caltech, turned hard-to-recycle PVC into polyalphaolefin lubricant at 70°C.
- What happened: Greg Liu’s group puts PVC — pipe, window frame, even a toy frog — in solvent with aluminum trichloride and alpha-olefins, heats it three hours at 158°F, and pulls a thick oil. Nature published the chemistry Aug. 5; ScienceDaily pushed it Aug. 29, which is why it is in this window. - One-line: A restorer brought up an IBM 7496 Executive Workstation — late-1980s prototype with the monitor fixed to a swiveling case — and gave it a hard disk.
- What happened: Hackaday’s Aug. 13 piece (recirculated in this week’s TWIAR headlines) follows [Epictronics] through a dead 7496: odd internals versus a normal PS/2, a sick 720K floppy, MCGA graphics.
